JP-153 is a promising small-molecule inhibitor that targets the c-Myc-TRRAP protein-protein interaction and has shown potent anti-tumor activity in preclinical studies. Its mechanism of action provides a potential strategy for cancer therapy that could be used in combination with other anti-cancer agents.
The c-Myc oncogene is a transcription factor that regulates a variety of cellular processes, including cell proliferation, apoptosis, and differentiation. Its overexpression is a common feature of many tumors, making it an attractive target for cancer therapy. TRRAP is a co-activator of c-Myc that promotes its transcriptional activity by facilitating its binding to chromatin. The interaction between c-Myc and TRRAP is critical for the maintenance of tumor growth and survival, and therefore inhibiting it has long been considered a potential strategy for cancer treatment.
JP-153 works by disrupting the interaction between c-Myc and TRRAP, thereby inhibiting the transcriptional activity of c-Myc and leading to cell cycle arrest and apoptosis. It does so by binding to the TRRAP domain that interacts with c-Myc, preventing the two proteins from binding to each other. This results in the downregulation of c-Myc target genes and inhibition of cancer cell growth.
